The isolated blood-perfused lung preparation is widely used to visualize and define signaling in single microvessels. By coupling this preparation with real time imaging, it becomes feasible to determine permeability changes in individual pulmonary microvessels. Herein we describe steps to isolate rat lungs and perfuse them with autologous blood. Then, we outline steps to infuse fluorophores or agents via a microcatheter into a small lung region. Using these procedures described, we determined permeability increases in rat lung microvessels in response to infusions of bacterial lipopolysaccharide. The data revealed that lipopolysaccharide increased fluid leak across both venular and capillary microvessel segments. Thus, this method makes it possible to compare permeability responses among vascular segments and thus, define any heterogeneity in the response. While commonly used methods to define lung permeability require postprocessing of lung tissue samples, the use of real time imaging obviates this requirement as evident from the present method. Thus, the isolated lung preparation combined with real time imaging offers several advantages over traditional methods to determine lung microvascular permeability, yet is a straightforward method to develop and implement.  相似文献

Pallister-Killian syndrome (PKS) is a rare sporadic genetic disorder usually caused by mosaicism of an extra isochromosome of 12p (i(12p)). This retrospective study analysed the prenatal ultrasound manifestations and molecular and cytogenetic results of five PKS foetuses. Samples of amniotic fluid and/or cord blood, skin biopsy and placenta were collected. Conventional karyotyping and single nucleotide polymorphism array (SNP array) were performed on all the amniotic fluid or cord blood samples. Copy number variants sequencing (CNV-seq) and fluorescence in situ hybridization (FISH) were also used for the validation for one foetus. All the five foetuses were from pregnancies with advanced parental age. Two foetuses involved structural abnormalities and one foetus had only soft markers, all of which included increased nuchal translucency. The rest two foetuses had normal ultrasounds in the second trimester, which has rarely been reported before. The karyotype revealed typical i(12p) in four cases and a small supernumerary marker chromosome consisting of 12p and 20p in the remaining one case. The proportion of cells with i(12p) ranged from 0 to 100% in cultural cells, while SNP array results suggested 2−4 copies of 12p. For one foetus, metaphase FISH showed normal results, but the interphase FISH suggested cell lines with two, three and four copies of 12p in the amniotic fluid. Advanced parental age may be an important risk factor for PKS, and there were no typical ultrasound manifestations related to PKS. A combination of karyotype analysis and molecular diagnosis is an effective method for the diagnosis of PKS.  相似文献

Coastal-ridge plains are progradational landforms composed of elevated ridges and low-lying swales. The transitions between ridges and swales are steep, promoting dynamic shifts between xeric and mesic systems. Two understory plants that co-occur in ridge plains of North American mid-Atlantic maritime forests are Sabal minor and Ilex vomitoria. As coastal-ridge plains foster varying amounts of surface and sub-surface water driven largely by topography, the purpose of this study was to evaluate plant–water relations and chlorophyll a fluorescence in these two species. Ridge plants had lower leaf- and xylem-water potentials, lower osmotic potential (I. vomitoria), and lower symplastic water content (S. minor). Although there were no differences in potential- and effective-quantum yields, there were decreases in fluorescence performance index for ridge I. vomitoria and swale S. minor. While the data support potential water-stress conditions in ridge plants of both species, the data also suggest that I. vomitoria and S. minor use different physiological processes to tolerate hydrologically dynamic ridge–swale maritime forests.  相似文献

In the course of study on the mechanism of the tartaric acid formation from 5-ketogluconic acid, a new intermediary substance with mauve color to Abdel-Akhel and Smith’s reagent was isolated from intact cell culture liquid. The chemical structure of this substance was determined as 1,2-dihydroxyethyl hydrogen L(+) tartrate from the results of hydrolysis experiments and from the identifications of the constituents of the molecule, and named “pretaric acid.” Tartaric acid was evidently produced from pretaric acid by intact cell culture. Clearly, then, pretaric acid appears to be an intermediate in the formation of tartaric acid from 5-ketogluconic acid. The authors assumed that in the formation of pretaric acid from 5-ketogluconic acid, a Baeyer-Villiger type oxidation occurred.  相似文献

Vancomycin-resistant enterococci (VRE) infections have been linked to increased mortality and costs. A new model of a VRE-infested intensive care unit (ICU) is introduced. It incorporates critical features including the difference between colonization and infection, the role of special preventive care treatment cycles, fitness cost, and antibiotic use. Five patient stages are considered: susceptible, colonized with and without special preventive care, and infected with and without treatment. Parameter ranges are determined representing different ICUs and incorporated to numerically simulate the model. Basic reproductive number of the infection is derived and the impacts of the parameters are analysed. Strategies to minimize VRE infections and outbreak risk are explored with a focus on efficient and simultaneous control of critical parameters. In particular, threshold values of the level of special preventive care and ICU compliance rate are given to achieve desired goals under various constraints.  相似文献

α-Tocopherol performs an antioxidant role in biological membranes by acting as a one-electron reductant. In micellar solutions it has been observed by pulse radiolysis that the micellar charge has a pronounced effect on the rate constant for repair of organic free radicals by α-tocopherol. The interactions between α-tocopherol and model bilayer lipid membranes have been studied by fluorescence spectroscopy. Quencing of α-tocopherol fluorescence by acrylamide and some n-doxyl stearates shows the transverse distribution of α-tocopherol in membranes to be affected by the physical state of the membrane lipids and by the salt concentration in the aqueous phase. Time-resolved fluorescence depolarization measurements, with a diphenylhexatriene-phospholipid conjugate as probe. demonstrate an increase in the bilayer order parameter on incorporation of α-tocopherol into a membrane  相似文献

Two novel, non-destructive assays were developed to evaluate contaminant-induced lipid peroxidation (thiobarbituric acid-reacting substances, TBARS, levels) and haem biosynthesis disruption (porphyrin excretion) in decapod crabs. A laboratory experiment was conducted whereby pie-crust crabs (Cancer novaezelandiae) were fed cockles (Austrovenus stutchburyi) collected from a contaminated and reference site and TBARS levels and porphyrin excretion determined using fluorometric analysis in urine samples. Pyrene metabolite levels were also measured in the same urine samples to assess polycyclic aromatic hydrocarbon (PAH) exposure. Contaminant-exposed crabs exhibited elevated urinary TBARS and porphyrin levels and a strong correlation was found between these two assays and the urinary pyrene metabolite concentrations. However, there was large within-treatment variability, which precluded a clear separation between the control and the impacted group. Nevertheless, consistency in the direction of the response shows that the biomarkers reflect pollutant levels and validates the use of these simple techniques from human medicine for environmental assessments.  相似文献

Photosynthetic characteristics, specific leaf mass, chlorophyll and total leaf nitrogen concentrations of four herbaceous plants (Dicranopteris linearis, Hanguana malayana, Pentaphragma ellipticum, Tacca integrifolia) from nutrient-poor tropical forests showed that all these plants were well-adapted to their natural growth environments. No photoinhibition was observed even in the understorey plants. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

In the lab, we exposed three foliose lichen species, Lobaria pulmonaria, Parmelia sulcata and Xanthoria aureola, to 0, 0.01, 0.2, and 0.6 M NaCl in combinations with copper and zinc (0, 10, 100, 500 μM). High salt concentrations adversely affected the lichen membrane integrity as measured by conductivity methods, whereas the potential photosystem II efficiency (Fv/Fm) was tolerant. High light was necessary to reduce Fv/Fm in thalli exposed to salt, whereas high light did not aggravate the conductivity. The seashore species X. aureola was much more resistant to salt than the old forest species L. pulmonaria. With respect to Cu and Zn, used concentrations had no (P. sulcata, X. aureola) or small (L. pulmonaria) effects on Fv/Fm. However, both heavy metals substantially increased conductivity in all species, consistent with membrane damage. Thus, the conductivity method detected high salt, high copper and high zinc stress much more efficiently than did the chlorophyll fluorescence method. This suggests that membrane integrity of the mycobiont is more sensitive to salt and heavy metal stress than potential photosystem II efficiency of its autotrophic partners.  相似文献
